Evaluation of Cell Viability Test by Flowcytometric Assay

|
Identification of viability of cells is essential for obtaining accurate data in cell culture. Conventionally, vital dyes such as trypan blue, have been used for discrimination of dead cells from viable cells by light microscope. We evaluated the
use of
an alternative method of flowcytometric analysis of propidium iodide-stained cells, and compared its results with those of the conventional method. The correlation of the results of these two methods was good (r=0.94, p<0.05) and the coefficients
of
variation from 5 day refrigerated storage sample was 2.8% in flowcytometric method and 14.1% in manual method. The counting time of flowcytometric method was 45 seconds per sample. It is concluded that the automated method with flowcytometry is
superior
to manual trypean blue method in precision and more convenient and time saving for testing large number of samples.
